A Comparison of Impedance and Optical Method for Evaluation of ?Platelet Aggregation |

Abstract |
The platelet aggregation studies are usually performed by the optical method using platelet-rich=plasma (PRP). One of the limitations of optical method is the need for centrifugation and separation of platelets from other blood cells, which may modulate platelet behavior. The platelet aggregation also can be studied conveniently with the impedance method, introduced in 1980 by Cardinal and Flower, using both PRP and whole blood (WB). We have investigated platelet behavior by three different techniques ; an optical method using PRP and impedance methods using PRP and WB. Blood samples were obtained in fasting state from 32 healthy volunteers (age 23-41 years, male 23, female 9) who had not taken any drugs which may interfere in platelet behavior for two weeks. The collecting blood was mixed with 1/9 volume of 3.8% sodium citrate, and centrifuged at 160g for 10 minutes to prepare PRP and 1500g for 20 minutes to platelet-poor-plasma (PPP), respectively. The platelet aggregation was measured at 37oC within 2 hour after venipuncture by the optical method on a NBS hematracer 801 aggregometer and by the impedance medthod on a chronolog whole blood aggregometer 560VS simultaneously. The light transmittance in PRP incereased with dose dependent patterns when different final concentrations of ADP(1, 5, 10, 20 uM) and collagen (1, 2ug/ml) were used. These changes in impedance method paralled that of the optical method, not only in PRP but also in the corresponding whole blood. Whereas there was good correlation between the aggregation curve patterns and the final concentrations of aggregation inducing agents. The dose-response relationship for ADP and collagen was nearly same when comparing with the three different methods. Therefore we concluded that platelet aggregation can be studied conveniently with the impedance method in the more physiologic medium containing all blood components that might modulate platelet behavior.
